Apart from moral questions about inflicting pain and death on animals for the purpose of medical research, there are an increasing number of questions about the suitability of using laboratory animals for research into human diseases and drug testing.<\/p>\n<p>There are many examples clearly showing that animal models are not predictive of human health. Recently, a group of scientists wrote an open letter in <a href=\"http:\/\/linkinghub.elsevier.com\/retrieve\/pii\/S0140673611608027\">The Lancet<\/a> (addressed to the UK prime minister and health secretary) pointing out that more than 90% of new drugs fail in clinical trials.<\/p>\n<p>The authors of the letter say part of the reason for this is the reliance on animal testing to predict drug behaviour in people. They claim that animal tests do this with the same probability as the toss of a coin. And they suggest that animal research be replaced with promising new technologies that increase clinical predictability as well as improving efficiency and cost.<\/p>\n<p>Other researchers have voiced similar concerns. An expert scientific group on phase one (screening for safety) clinical trials in the United Kingdom, for instance, <a href=\"http:\/\/www.dh.gov.uk\/en\/Publicationsandstatistics\/Publications\/PublicationsPolicyAndGuidance\/DH_063117\">concluded<\/a> that pre-clinical animal studies may not reproduce the intended pharmacological effect, may give misleading results and, most significantly, may not reflect toxicity in humans.<\/p>\n<figure class=\"align-left\"><img src=\"https:\/\/62e528761d0685343e1c-f3d1b99a743ffa4142d9d7f1978d9686.ssl.cf2.rackcdn.com\/files\/13828\/width237\/4qyw22x2-1343971013.jpg\" alt=\"\" \/><figcaption><span class=\"caption\">Penicillin may not have seen the light of day of it had been tested in guinea pigs because it\u2019s fatal to them.<\/span><br \/>\n<span class=\"attribution\"><span class=\"source\">Linda Bartlett<\/span><\/span><\/figcaption><\/figure>\n<p>While humans and other animals have biological and physiological similarities, differences at the molecular and cellular levels can result in different responses to drugs.<\/p>\n<h2>Animal testing gone wrong<\/h2>\n<p>The thalidomide tragedy of the late 1950s and early 1960s is a well-known example for the inappropriateness of animal research. <a href=\"http:\/\/www.scienceboard.net\/community\/perspectives.245.html\">Thalidomide<\/a> had been tested on several animal species and found to be safe, even in large doses. Yet thousands of children worldwide were born with missing or shortened limbs after their mothers took the drug for morning sickness.<\/p>\n<p>The website of <a href=\"http:\/\/www.humaneresearch.org.au\/dangerous-drugs\">Humane Research Australia<\/a> lists over 60 drugs, including thalidomide, that have been \u201csuccessfully\u201d tested on animals, but are toxic to humans. The site notes that the list is not exhaustive.<\/p>\n<p>In contrast, <a href=\"http:\/\/www.afma-curedisease.org\/pdf\/penicillin.pdf\">penicillin<\/a> may not have seen the light of day if it had been tested on guinea pigs or rats \u2013 it kills guinea pigs and produces birth defects in rats. In fact, this antibiotic <a href=\"http:\/\/www.opposingviews.com\/i\/society\/animal-rights\/no-responsibility\">stayed on the shelf<\/a> for over a decade because it was not effective in the rabbits on which it had been tested.<\/p>\n<p>There are many other <a href=\"http:\/\/www.safermedicines.org\/quotes\/cancer.shtml\">examples<\/a> showing animal testing to have very poor predictive value for human diseases and toxicity. But animals are still used in laboratories all over the world to test the safety, toxicity and effectiveness of drugs. In fact, (and rather paradoxically) animal testing was made mandatory by drug regulators after the thalidomide tragedy. During the more than four decades since, it has become clear that animal tests fail to accurately predict human responses. And now, new testing methods are available.<\/p>\n<p>In their book <a href=\"http:\/\/www.amazon.com\/Sacred-Cows-Golden-Geese-Experiments\/dp\/0826412262\">\u201cSacred cows and golden geese\u201d<\/a> Ray Greek and Jean Swingle Greek write that they could not find instances of advances that could be credited to the animal model despite searches of medical literature. Most drug advances are due to clinical observations by doctors and nurses on their human patients. But many clinical discoveries, discoveries by serendipity or other non-animal based discoveries were later \u201cvalidated\u201d in animals.<\/p>\n<figure class=\"align-center\"><img src=\"https:\/\/62e528761d0685343e1c-f3d1b99a743ffa4142d9d7f1978d9686.ssl.cf2.rackcdn.com\/files\/13829\/width668\/jvtjkyqk-1343971237.jpg\" alt=\"\" \/><figcaption><span class=\"caption\">Most drug advances are due to clinical observations are later \u2018validated\u2019 in animals.<\/span><br \/>\n<span class=\"attribution\"><span class=\"source\">Armin K\u00fcbelbeck<\/span><\/span><\/figcaption><\/figure>\n<h2>Emerging alternatives<\/h2>\n<p>Humane Research Australia provides a <a href=\"http:\/\/www.humaneresearch.org.au\/fact-sheets\">fact sheet<\/a> with a range of alternatives to animal-based research. They include:<\/p>\n<ul>\n<li>a greater focus on prevention;<\/li>\n<li>epidemiology (the study of human populations);<\/li>\n<li>autopsies;<\/li>\n<li>in vitro research (using tissue and cell cultures); and<\/li>\n<li>computer modelling.<\/li>\n<\/ul>\n<p>Genomics (the study of the chromosomes of an organism), proteomics (study of proteins, particularly their structures and functions), nanotechnology, phage display (a method of quickly evaluating a huge range of potentially useful antibodies and then producing large quantities of the selected ones) and <a href=\"http:\/\/news.stanford.edu\/news\/2006\/january18\/fluidics-011806.html\">microfluidic chips<\/a> (silicon microelectronics), are all new technology alternatives to animal testing that are worth considering.<\/p>\n<p>Human organs on a chip, <a href=\"http:\/\/www.caseyresearch.com\/articles\/animal-testing-about-become-obsolete\">described<\/a> as \u201ccutting tiny grooves into silicon and plastic substrates, introducing living cells into the spaces, and hoping the end result will mimic a particular biological system, like a human organ\u201d, are <a href=\"http:\/\/www.caseyresearch.com\/articles\/animal-testing-about-become-obsolete\">reportedly<\/a> close to practical application.<\/p>\n<p>The aim of this research is not to make replacement organs for transplant, but to replicate an organ\u2019s functions to make the chips useful in testing substances for toxic and therapeutic effects. Organs on a chip have so far been developed for the human <a href=\"http:\/\/medgadget.com\/2010\/07\/lung_on_a_chip.html\">lung<\/a> and <a href=\"http:\/\/medgadget.com\/2012\/03\/human-intestine-becomes-latest-organ-to-be-put-on-a-chip.html\">intestine<\/a>. And this technology could help <a href=\"http:\/\/blogs.fda.gov\/fdavoice\/index.php\/tag\/human-on-a-chip\/\">shorten the time<\/a> it takes for new treatments to move to human testing and approval for general use.<\/p>\n<figure class=\"align-right\"><img src=\"https:\/\/62e528761d0685343e1c-f3d1b99a743ffa4142d9d7f1978d9686.ssl.cf2.rackcdn.com\/files\/13822\/width237\/6hz4j5m6-1343968618.jpg\" alt=\"\" \/><figcaption><span class=\"caption\">A monkey photographed at an animal testing facility in the United States.<\/span><br \/>\n<span class=\"attribution\"><span class=\"source\">SlimVirgin\/Wikimedia Commons<\/span><\/span><\/figcaption><\/figure>\n<h2>Moving away from animal testing<\/h2>\n<p>Early this year, <a href=\"http:\/\/www.abc.net.au\/7.30\/content\/2012\/s3411750.htm\">ABC\u2019s 7.30 Report<\/a> looked at a supercomputer in Melbourne. <a href=\"http:\/\/www.vlsci.org.au\/researcher\/prtaylor\">Professor Peter Taylor<\/a>, the Director of the <a href=\"http:\/\/www.vlsci.org.au\/\">Victorian Life Science Computation Initiative<\/a> at the University of Melbourne said, \u201cOne could imagine that if our ability to model how, say, drugs interact with their targets and how they affect an organism as a whole, then we could basically avoid any sort of animal testing. We could do it all in silica, if you like, in the computer, which would certainly be a great benefit. I don\u2019t think there\u2019s anybody who would quarrel with trying to do that.\u201d<\/p>\n<p>In the United States, the <a href=\"http:\/\/www.nytimes.com\/2011\/12\/16\/science\/chimps-in-medical-research.html\">National Institutes of Health<\/a> recently suspended all new grants for biomedical and behavioural research on chimpanzees after an expert committee <a href=\"http:\/\/iom.edu\/Reports\/2011\/Chimpanzees-in-Biomedical-and-Behavioral-Research-Assessing-the-Necessity.aspx\">concluded<\/a> that most such research was unnecessary. And the US <a href=\"http:\/\/dels.nas.edu\/Materials\/Report-In-Brief\/4640-Toxicity-Testing\">National Research Council<\/a> recommends animal-based tests be replaced as soon as possible with human cell-based assays, in silico (computer) models, and an increased emphasis on epidemiology.<\/p>\n<p>At least one pharmaceutical company has already ceased its animal research program. Denmark-based Novo Nordisk <a href=\"http:\/\/www.pharmaceutical-technology.com\/news\/newsnovo-nordisk-to-end-animal-testing\/\">stopped<\/a> using animals for testing its products in November 2011.<\/p>\n<p>New medicines should be safe and effective, but animal testing is a flawed model.
